The Impact of the Circadian Clock on Skin Physiology and Cancer Development

Abstract: Skin cancers are growing in incidence worldwide and are primarily caused by exposures to ultraviolet (UV) wavelengths of sunlight. UV radiation induces the formation of photoproducts and other lesions in DNA that if not removed by DNA repair may lead to mutagenesis and carcinogenesis. Though the factors that cause skin carcinogenesis are reasonably well understood, studies over the past 10–15 years have linked the timing of UV exposure to DNA repair and skin carcinogenesis and implicate a role for the body’s c… Show more

“…There is great interest in improving the effectiveness of sunscreens to protect vulnerable individuals from mutagenesis and carcinogenesis associated with exposures to solar UV radiation, including through the use of natural products and the application of sunscreens containing DNA repair enzymes (1,2,30). Because the natural product NOB has been shown to enhance the transcriptional output of the circadian transcriptional machinery (7), which is known to regulate UV photoproduct removal and other UVB DNA damage responses (14,15,17), we investigated value of NOB in keratinocyte responses to UV radiation. Though NOB protected both HaCaT and telomerase-immortalized diploid keratinocytes from UVB radiation, we found this mechanism to be independent of DNA repair and the core circadian clock protein BMAL1.…”

“…Daily variation in gene expression have been reported in the epidermis and dermis, as well as in cutaneous and SATs and dermal hair follicles [ 38–40 ]. At the cellular level epidermal keratinocytes, hair follicle cells, dermal fibroblasts and dermal macrophages all generate rhythms on a daily timescale, as do subcutaneous adipocytes [ 41 ].…”

“…The circadian rhythm, or circadian clock (CC), is a hierarchically cyclic system that regulates the daily oscillations of physiological processes and can respond to external environment changes to maintain internal homeostasis [ 1 ]. Because some of these outside shifts are caused by the 24-h rotation of the Earth, and so are expected to occur approximately at the same time every day, the circadian regulation allows the human organism to anticipate these changes and synchronize them with inner physiology [ 2 ].…”
