The inhibitory effect of BSP‐A1/‐A2 on protein kinase C and tyrosine protein kinase

Abstract: Bovine seminal plasma contains a group of similar proteins, namely BSP-A1, BSP-A2, BSP-A3, and BSP-30-kDa (collectively called BSP proteins), and they are secreted by the seminal vesicles. In our study, we purified the BSP-A1/-A2 through affinity chromatography and found for the first time that BSP-A1/-A2 can inhibit the activity of protein kinase C (PKC) and tyrosine protein kinase (TPK). The inhibition was dose dependent. When the PKC and TPK activities are expressed as the logarithm of percentage activity t… Show more

“…Indeed, the stimulation of BSPs for capacitation is dependent on exposure time and presence of heparin or high‐density lipoprotein or glycosaminoglycan present in the oviductal fluid (Manjunath and Therien, 2002). On the contrary, Yu et al (2003) have proposed that PDC‐109 also prevents premature acrosome reaction of spermatozoa in the female reproductive tract by inhibiting protein kinase C activity. BSP proteins exert both beneficial and detrimental effects, which are important in maintaining spermatozoa in an appropriate state to facilitate fertilization in the female reproductive tract.…”

“…The major bovine SPPs are BSP-A1/A2, BSP-A3 and BSP-30 kDa which play substantial roles in fertility by maintaining sperm in an appropriate state in the female genital tract until the oocyte reaches the site of fertilisation (Yu et al, 2003;Gwathmey et al, 2006). Our results are partially consistent with these findings because protein bands of 15 and 13.5 kDa which might correspond to BSP-A1 or BSP-A3 and BSP-A2, respectively were the most abundant proteins in these fertile bulls.…”

“…Moreover, two additional functions were proposed for BSPs. The first one is the activity of protein kinase C (PKC) and of tyrosine protein kinase (TPK) inhibited in vitro by BSP-A1/A2 (Yu et al, 2003). It was proposed that the inhibition of PKC may serve to prevent the premature acrosome reaction of sperm in the female geni-tal tract.…”

“…Abundant low molecular weight bovine seminal plasma proteins were replaced during cryopreservation by high molecular weight extender-derived chicken structural proteins. Major bovine seminal plasma proteins PDC-109 (BSP-A1/A2), BSP-A3 and BSP-30 kDa play important roles in fertility by maintaining sperm in an appropriate state in the female tract until the oocyte reaches the site of fertilization [10][11][12][13]. It is unclear whether the effect of processing that decreased the abundance of these BSP proteins by more than half would render them ineffective in preventing premature capacitation and acrosome reaction of sperm from normal fertility bulls in the female tract.…”
