The LAT Story: A Tale of Cooperativity, Coordination, and Choreography

Abstract: The adapter molecule LAT is a nucleating site for multiprotein signaling complexes that are vital for the function and differentiation of T cells. Extensive investigation of LAT in multiple experimental systems has led to an integrated understanding of the formation, composition, regulation, dynamic movement, and function of LAT-nucleated signaling complexes. This review discusses interactions of signaling molecules that bind directly or indirectly to LAT and the role of cooperativity in stabilizing LAT-nuclea… Show more

“…Nef affects the subcellular localization of the adaptor protein LAT Formation of SLP-76 MCs critically depends on the adaptor protein LAT (11,54,55). In search for a mechanism by which Nef prevents the formation of SLP-76 MCs, we therefore analyzed next whether the viral protein affects the subcellular localization of LAT using Jurkat E6.1 cells stably expressing LAT.GFP that had been FACS sorted for medium levels of LAT.GFP expression.…”

“…Active ZAP70 subsequently phosphorylates two critically important adaptor proteins, linker of activated T cells (LAT) and Src homology-2 domain-containing leukocyte protein of 76 kDa (SLP-76) (10). LAT facilitates the formation of several multiprotein complexes and acts as nucleation center for T cell signal transduction (11). Upon phosphorylation at Tyr 191 by ZAP70 (12), LAT recruits SLP-76 into MCs via an additional adaptor protein, GADS (13).…”

“…They can also appear as membrane-associated structures or may even be combined with catalytic domains, as in the case of the transmembrane receptor kinases [84,87]. The TCR component LAT and the EGF receptor are representatives and well-studied examples of two major types of membrane-associated signal complex assembly sites (see [88] and [89] for details on their architectures and interactions).…”

“…LAT partitions into two pools, one at the plasma membrane and a second, located in intracellular compartments ( Figure 2). Upon phosphorylation, plasma membrane localized LAT redirects the intracellular LAT pool to the nascent IS [30,31]. The initial TCR signal is amplified after CD3 and LAT signalosomes localize to their corresponding microclusters at the plasma membrane [32], as well as after their recycling and sorting from intracellular stores.…”

“…Additionally in CD8+ CTLs, suboptimal activation of the Centrosome polarization at the IS also allows trafficking of the membrane adaptor protein Linker for activation of T cells (LAT) to the IS. LAT is phosphorylated at Y132 by ZAP70, allowing PLCγ1 binding and activation [30]. LAT partitions into two pools, one at the plasma membrane and a second, located in intracellular compartments ( Figure 2).…”
