The linear 20 kb mitochondrial genome of Pandorina morum (Volvocaceae, Chlorophyta)

Abstract: A physical restriction map of the mitochondrial genome from one clone (TCC 854) of the sexually isolated populations (syngens) of the morphologically uniform species Pandorina morum Bory has been constructed using restriction endonucleases Ava I, Bam HI, Bgl II, Eco RI, Kpn I, and Pst I. The 20 kb linear genome can easily be separated from plastid DNA, nuclear satellite rDNA, and main band (nuclear) DNA on a Hoechst/CsCl buoyant density gradient. The Pandorina mitochondrial DNA shows sufficient similarity to t… Show more

“…However, m t D N A of Chlamydomonas reinhardtii (Volvocales) is much smaller (15 kb [36]) and linear. Similar results were obtained for another species of the Vovocales, Pandorina morum, which was shown to possess a linear mitochondrial genome of about 20 kb [33]. For the chromophyte Olisthodiscus luteus a mitochondrial genome of 50 kb with circular shape has been demonstrated [ 1 ].…”

“…Moreover, in addition to C. reinhardtii , other Reinhardtinia algae are now known to have linear mitochondrial genomes (fig. 1 A ), such as Chlamydomonas asymmetrica (Laflamme and Lee 2003), the colonial volvocine Pandorina morum (Moore and Coleman 1989), and all known members of the nonphotosynthetic genus Polytomella (Smith, Hua, et al. 2013).…”

“…A linear mt genome flanked by inverted repeats has also been reported for the green alga Pandorina morum (Moore and Coleman 1989). In a phylogenetic analysis based on molecular, i.e., nuclear-encoded SSU and LSU rRNA sequences, and organismal data, this multicellular species turned out to be more closely related to C. reinhardtii and C. smithii than to C. eugametos and C. moewusii (Buchheim et al 1994).…”
