The <i>Drosophila Neprilysin 4</i> gene is essential for sperm function following sperm transfer to females

Abstract: Sperm are modified substantially in passing through both the male and the female reproductive tracts, only thereafter becoming functionally competent to fertilize eggs. Drosophila sperm become motile in the seminal vesicle; after ejaculation, they interact with seminal fluid proteins and undergo biochemical changes on their surface while they are stored in the female sperm storage organs. However, the molecular mechanisms underlying these maturation processes remain largely unknown. Here, we focused on Drosoph… Show more

“…Fruitless , a male sexual determination gene [ 28 ], is only expressed in the TE. Additionally, the mRNA of boule and neprilysin-4, important for spermatogenesis [ 29 , 30 ], nanos and ovo , essential for oogenesis [ 31 , 32 ], dupd1 , the encoding products of which belong to the seminal fluid peptides [ 33 ], and lysozyme 1, related to the antimicrobials of FAG [ 34 ], are specifically expressed in the TE, OV, MAG, and FAG, respectively ( Figure 1 E). This suggests that the DEG analysis is relatively accurate.…”

“…Since no previous studies were available on Ceratitis capitata , we first selected seven candidate genes to identify suitable targets for robust RNAi‐mediated control of this insect. The selection was based on previous successful reports of increased mortality in other dipteran and insect species: Snf7 , 44,56 RyR , 57,58 Nep4 , 59,60 IAP2 , 58 ATPsynbeta , 61,62 V‐ATPase , 63–65 and RPS13 45,62,66 . Target regions between 400 and 500 nt in the coding sequences of these genes were selected, taking into account their essentiality and specificity.…”

“…Nep4 , a drosophila ortholog of mammalian Mmel1 , encodes a metalloprotease expressed in male gonads ( 136 ). Nep4 mutant males are infertile; mutant sperm are quickly discarded by females ( 30 ). When Dcp-1 encoding a cysteine protease was ablated in their germline, the resulting females were infertile because of defective oogenesis ( 31 ).…”
