The molecular architecture of glucose‐1‐phosphate uridylyltransferase

Abstract: Glucose-1-phosphate uridylyltransferase, also referred to as UDP-glucose pyrophosphorylase or UGPase, catalyzes the formation of UDP-glucose from glucose-1-phosphate and UTP. Not surprisingly, given the central role of UDP-glucose in glycogen synthesis and in the production of glycolipids, glycoproteins, and proteoglycans, the enzyme is ubiquitous in nature. Interestingly, however, the prokaryotic and eukaryotic forms of the enzyme are unrelated in amino acid sequence and structure. Here we describe the clonin… Show more

“…As highlighted by STD NMR spectroscopic studies, interactions of the uridine moiety of nucleotide sugars or UTP with LmjUSP are similar to those observed with LmjUGP and play a significant role in substrate binding. This leading role of the nucleotide moiety is observed in many enzymes involved in glycosylation, for example UDP-galactopyranose mutase (35), sialyltransferases (36), and pyrophosphorylases from E. coli (37), and might even hold true for nucleotide sugar transporters (38).…”

“…Beside the structures of GMP members, the structure most closely related to that of TmGMP is that of UDPGlc PPase from Corynebacterium glutamicum (CgUGP) (36,37). Most of the structural elements of CgUGP are similar to those of TmGMP, except that the CgUGP structure lacks TmGMP helices ␣7 and ␣8 and comprises two additional helices inserted in the ␣2-␤3 loop.…”

“…This enzyme synthesizes UDP-glucose from glucose-1-phosphate. UDP-glucose has been reported to be involved in galactose utilization, in glycogen synthesis, and in the synthesis of various carbohydrate moieties, such as glycolipids, glycoproteins, and proteoglycans (33). Interestingly, yfgC codes for a putative Zndependent protease, and although its function is unknown, the chemical and genetic data suggest that it also plays a role in outer membrane protein biogenesis (34).…”
