The maintenance of genome integrity is an essential trait to the successful transmission of genetic information. In animal germ cells, piRNAs guide PIWI proteins to silence transposable elements (TEs) in order to maintain genome integrity. In insects, most TE silencing in the germline is achieved by secondary piRNAs that are produced by a feed-forward loop (the pingpong cycle), which requires the piRNA-directed cleavage of two types of RNAs: mRNAs of functional euchromatic TEs and heterochromatic transcripts that contain defective TE sequences. The first cleavage that initiates such an amplification loop remains poorly understood. Taking advantage of the existence of strains that are devoid of functional copies of the LINE-like I-element, we report here that in such Drosophila ovaries, the initiation of a ping-pong cycle is exclusively achieved by secondary I-element piRNAs that are produced in the ovary and deposited in the embryonic germline. This unusual secondary piRNA biogenesis, detected in the absence of functional I-element copies, results from the processing of sense and antisense transcripts of several different defective I-element. Once acquired, for instance after ancestor aging, this capacity to produce heterochromatic-only secondary piRNAs is partially transmitted through generations via maternal piRNAs. Furthermore, such piRNAs acting as ping-pong initiators in a chromatin-independent manner confer to the progeny a high capacity to repress the I-element mobility. Our study explains, at the molecular level, the basis for epigenetic memory of maternal immunity that protects females from hybrid dysgenesis caused by transposition of paternally inherited functional I-element.

Using data from in situ hybridisation of the giant chromosomes from salivary glands, mdg-1, I, copia, and P mobile element polymorphism was studied in 17 highly-inbred lines of Drosophila melanogaster and in the expected hybrids obtained from the theoretically-crossed inbred lines. The mean copy numbers of each element on the inbred lines were close and equal to 16-8 for mdg-1, 17•5 for I, 178 for copia and 186 for P (these values included the insertions in the centromeric regions). The P element differed from the other three for variance in copy number and distribution of the number of insertion site occurrences. A low frequency of X-linked copia element insertions (as compared with frequency of insertions in the other chromosomes) was reported, suggesting that natural selection acts against insertional deleterious mutations of this element. Such a low frequency of X-linked insertions was not observed for the other three elements. The mdg-1, I and copia element copy numbers were positively correlated, thus leading to lines with high or low copy number. No correlation was observed between the P element copy number and the numbers of mdg-1, I and copia elements. Moreover, a genomic control of copy number was detected for mdg-1, I and copia, but not for P. The number of labelling sites, the number of copies, the number of heterozygous insertion sites, and various indices of genetic variability were estimated from the expected hybrids obtained from the theoretically-crossed inbred lines; the calculated numbers of labelling sites for mdg-1 and I did not differ from those obtained directly from the initial wild-derived population (we had no estimate of the number of labelling sites available in this population for copia and P). An average proportion of heterozygous sites of 086, 092, 0•90 and 094, and an average number of copies of 33, 35, 38 and 37 were found in the theoretical hybrids for the mdg-1, I, copia and P elements, respectively.

Revisiting horizontal transfer of transposable elements in Drosophila

Horizontal transfer (HT), defined as the transfer of genetic material between species, is considered to be an essential step in the 'life cycle' of transposable elements. We present a broad overview of suspected cases of HT of transposable elements in Drosophila. Hundred-one putative events of HT have been proposed in Drosophila for 21 different elements (5.0% refer to non-long terminal repeat (LTR) retrotransposons, 42.6% to LTR retrotransposons and 52.4% to DNA transposons). We discuss the methods used to infer HT, their limits and the putative vectors of transposable elements. We outline all the alternative hypotheses and ask how we can be almost certain that phylogenetic inconsistencies are due to HT.
