The Molecular Pathogenesis of Tumor-Suppressive miR-486-5p and miR-486-3p Target Genes: GINS4 Facilitates Aggressiveness in Lung Adenocarcinoma

Abstract: The involvement of passenger strands of miRNAs in the molecular pathogenesis of human cancers is a recent concept in miRNA research, and it will broaden our understanding of the molecular mechanisms of miRNA-mediated cancer. The analysis of our miRNA signature of LUAD revealed that both strands of pre-miR-486 (miR-486-5p and miR-486-3p) were downregulated in LUAD tissues. Ectopic expression of both miRNAs induced cell cycle arrest in LUAD cells, suggesting both strands of miRNAs derived from pre-miR-486 were t… Show more

“…However, LAC and PCT are susceptible to other factors and need dynamic monitoring to predict the prognosis of patients with SCAP. Previously, miR-486-5p has been reported to participate in the development of lung adenocarcinoma and pulmonary arterial hypertension [38][39][40]. Additionally, miR-486-5p was found to link idiopathic pulmonary fibrosis and COVID-19 predicting the development of fibrosis in post-COVID-19 patients [12,13].…”

“…We recently created a miRNA expression signature based on LUAD clinical specimens using RNA sequencing [10]. Using this signature, we selected miRNAs with suppressed expression in LUAD tissues.…”

“…Small RNAs (miRNAs and siRNAs) were transfected into LUAD cell lines, and cell proliferation, migration, and invasion were evaluated and compared with those of control cells. The transfection procedures of miRNAs and siRNAs were described in our previous studies [10,[15][16][17]. All miRNA precursors were transfected at 10 nM, and all siRNAs were transfected at 5 nM into A549 and H1299 cell lines using RNAiMAX (Invitrogen, Carlsbad, CA, USA).…”
