The N-rasoncogene assigned to the short arm of human chromosome 1

Abstract: The human N-ras oncogene, isolated from the HL-60 promyelocytic leukemia cell line, is distantly related to viral oncogenes of Kirsten and Harvey sarcoma viruses. We have determined its chromosomal location by Southern blot analysis of DNAs from 37 human x rodent hybrid cell lines derived from 8 different human donors, some of whom carried balanced rearrangements of chromosome 1. The results indicate that the N-ras oncogene (RASN) is localized on the proximal part of the short arm of human chromosome 1, in reg… Show more

“…Restriction-endonuclease digested DNA samples were subjected to electrophoresis in 0.7-1% agarose gels and transferred to nitrocellulose as described (4,5,16). Prehybridization, hybridization and washing of filters under stringent conditions of 65°C in 0.1% SDS, 0.1 x SSC (1 x SSC is 0.15 M NaCl, 15 mM sodium citrate), and the 32P-labeling of DNA have been detailed (4,5,16). RNA Isolation and Blot Hybridization Total cellular RNA was prepared with guanidine-hydrochloride (17).…”

“…We found a complex rearrangement of the short arm in two of the four copies of chromosome 11 present in this heteroploid cell line. In addition, numerous other chromosomes were rearranged (18). Furthermore, the t(3; 1 1) translocation found in tumor cells of a patient with hereditary renal cell carcinoma also involved a breakpoint in the short arm of 11 (region llpl5) (19).…”

“…Cytochalasins and colchicine derivatives have been used to study the roles of microfilaments and microtubules, respectively, in cell morphology, motility, and division (17)(18)(19)(20). These antimitotic drugs have been tested for indirect effects on the various IF systems with limited success (7,21).…”

“…D20S4, a 1.2-kb HindIII fragment derived from human chromosome 20 (4,5), was obtained in a recombinant pBR322 plasmid from Peter O'Connell (University of Utah). The probe recognizes a two-allele Msp I restriction fragment length polymorphism (RFLP).…”
