The NADPH Oxidase Isoform 1 Contributes to Angiotensin II-Mediated DNA Damage in the Kidney

Abstract: In higher concentrations, the blood pressure regulating hormone angiotensin II leads to vasoconstriction, hypertension, and oxidative stress by activating NADPH oxidases which are a major enzymatic source of reactive oxygen species (ROS). With the help of knockout animals, the impact of the three predominant NADPH oxidases present in the kidney, i.e., Nox1, Nox2 and Nox4 on angiotensin II-induced oxidative damage was studied. Male wildtype (WT) C57BL/6 mice, Nox1-, Nox2- and Nox4-deficient mice were eq… Show more

“…In addition to, TLR4 and PPAR-γ, Ang II is known to induce ROS via activation of NADPH oxidase mediated Nox-1 and p22phox in Ang II-induced DNA damage in-vivo and in-vitro models ( Wattanapitayakul et al, 2000 ; Sarr et al, 2006 ; Zimnol et al, 2020 ). Hyperactivation of Nox acts as a major source of ROS production in cardiac tissue, promotes apoptosis and increases oxidative stress via the MAPK pathway ( Wen et al, 2019 ).…”

“…The isoforms Nox 2 and 4 are widely expressed in cardiomyocytes ( Byrne et al, 2003 ; Murray et al, 2013 ). Nox 1 may play a major role in Ang II mediated impairment of kidney function by increasing oxidative stress ( Zimnol et al, 2020 ). Reactive oxygen species also increase intracellular calcium and activate NF-ҡB and activating protein-1.…”

“…Similarly, in hypertensive rats, Nox activation induced ROS production, while ROS production was decreased with knockout of the Nox1 gene [31]. Blood pressure elevation and vascular sclerosis induced by Ang II were signi cantly decreased in mice with knockout of the Nox1 gene [32]. In atherosclerotic mice, the neointima of the damaged carotid artery becomes thickened and is accompanied by Nox1 overexpression.…”
