The New Zealand obese mouse model of obesity insulin resistance and poor breeding performance: evaluation of ovarian structure and function

Abstract: Infertility, associated with oligo/anovulation, increased ovarian volume, numerous follicular cysts, and metabolic disturbances such as obesity and insulin resistance (IR) are characteristics common to polycystic ovary syndrome (PCOS), the most common endocrine disorder in women of reproductive age. Here, we show that New Zealand obese (NZO) mice display similar metabolic characteristics such as obesity, leptin insensitivity, glucose intolerance, and IR. Importantly, NZO mice are poor breeders; however, the me… Show more

“…This can be related to the reduced ovulatory rate or anovulation, explaining the poor fertility rate in this group. This finding is confirmed by Radavelli-Bagatini et al study (16). It seems these results may be due to increased serum estradiol concentration which can provide an inappropriate hormonal environment for developing follicles to be ovulated in very earliest stages of folliculogenesis (22).…”

“…All types of follicles were counted in each ovary. Then all follicles were classified as healthy (primary, preantral, antral and preovulatory) or atretic (16). All types of follicles, cysts, and corpus lutea in each ovary were counted by two persons within 9 representative sections at least 25 μm apart.…”

“…NZO is a polygenic model of obesity, insulin resistance, leptin insensitivity, glucose intolerance, and hyperinsulinemia. [6] Fed with carbohydrate-rich diet, those animals become quickly resistant to insulin and eventually diabetic. At the same time, they develop obesity.…”

“…Although they have normal leptin and leptin receptor genes, NZO mice exhibited a defect in leptin transport across the blood-brain barrier. Along with obesity, the NZO mouse exhibited insulin resistance and hyperinsulinemia, all of which are common metabolic abnormal characteristics of PCOS [99]. Furthermore, the mouse displayed dyslipidemia, hypercholesterolemia, and hypertension [100].…”
