The NF-κB Signaling Pathway, the Microbiota, and Gastrointestinal Tumorigenesis: Recent Advances

Abstract: Gastrointestinal (GI) cancers, especially gastric cancer and colorectal cancer (CRC), represent a major global health burden. A large population of microorganisms residing in the GI tract regulate physiological processes, such as the immune response, metabolic balance, and homeostasis. Accumulating evidence has revealed the alteration of microbial communities in GI tumorigenesis. Experimental studies in cell lines and animal models showed the functional roles and molecular mechanisms of several bacteria in GI … Show more

“…A similar mode of action has been seen in F. prausnitzii in different colitis models 62 , notably via the release of the microbial anti-inflammatory molecule (MAM). Although the NF-κB signaling pathway serves as a major line of defense against pathogens, it can have deleterious effects when overactivated due to the increased production of proinflammatory cytokines 72 . Consequently, it is important to determine which molecules help control pathway activation so that the development of inflammation can be halted.…”

“…NF-κB signaling is a central mediator of GC progression [ 24 ]. HOXA-AS3 is able to colocalize with NF-κB in the promoter region of certain genes, and can directly interact with this transcription factor in order to enhance its activation by regulating IκBα expression and p65 subunit K310 acetylation status [ 16 ].…”
