The nucleoprotein of severe fever with thrombocytopenia syndrome virus processes a stable hexameric ring to facilitate RNA encapsidation

Abstract: Severe fever with thrombocytopenia syndrome virus (SFTSV), a member of the Phlebovirus genus from the Bunyaviridae family endemic to China, is the causative agent of life-threatening severe fever with thrombocytopenia syndrome (SFTS), which features high fever and hemorrhage. Similar to other negative-sense RNA viruses, SFTSV encodes a nucleocapsid protein (NP) that is essential for viral replication. NP facilitates viral RNA encapsidation and is responsible for the formation of ribonucleoprotein complex. Howe… Show more

“…Additionally, hantavirus NP core also exhibits homology with NPs encoded by phleboviruses, including RVFV (PDB code 4H5 L) (11-13), SFTSV (PDB code 4J4X) (14,19), and Toscana virus (PDB code 4CSG) (58). The Z score between SNV NP core and the phlebovirus NP is approximately 5.8, and the RMSD of 148 aligned residues is 4.5 Å, suggesting that they have partial structural homology (Fig.…”

“…The RNP either further serves as an active template for RNA synthesis or is packaged into the virion (8)(9)(10). Since 2010, a series of structural studies, including studies on the structures of NPs encoded by phleboviruses (Rift Valley fever virus [RVFV] [11][12][13] and severe fever with thrombocytopenia syndrome virus [SFTSV] [14]), nairovirus (CrimeanCongo hemorrhagic fever virus [CCHFV]) (6,15,16), and orthobunyaviruses (Bunyamwera virus [BUNV], etc.) (17)(18)(19)(20)(21), has advanced the understanding of the structures and functions of bunyavirus NPs.…”

“…Using an ambisense coding strategy, the S segment encodes the nucleoprotein and a nonstructural protein (NSs). The nucleoprotein is a multifunctional protein that protects the viral genome, is involved in the replication/ transcription process, and has a role in virion assembly (3,4). The NSs protein of SFTSV has been shown to inhibit the type I interferon (IFN) response through sequestration of STAT proteins and other factors in viral replication complexes (5)(6)(7).…”

“…After years of NP structural investigation, several NSV nucleoprotein structures have been reported, such as Lassa fever virus (LAFV) (10,11), Crimean-Congo hemorrhagic fever virus (CCHFV) (12,13), influenza A virus (14), hantavirus (15), and SFTS virus (16) among segmented NSVs (sNSVs) and Borna disease virus (BDV) (17), vesicular stomatitis virus (VSV) (18,19), respiratory syncytial virus (RSV) (20), parainfluenza virus 5 (PIV-5) (21), and Nipah virus (NiV) (22) among nonsegmented NSVs. Despite the structural differences among these nucleoproteins, most of the structures share a typical bilobe fold consisting of N-and C-terminal domains, with a positive-charged RNA binding groove between the two lobes.…”
