The Organization and Duplication of Chromosomes as Revealed by Autoradiographic Studies Using Tritium-Labeled Thymidinee

Abstract: Information on the macromolecular organization of chromosomes and their mode of duplication has been difficult to obtain in spite of numerous attempts. One point of attack, long recognized but until recently unattainable, was the selective labeling of some component of the chromosome, the distribution of which could be seen in succeeding cell divisions. Reichard and Estborn' demonstrated that N15-labeled thymidine was a precursor of deoxyribonucleic acid (DNA) and that it was not diverted to the synthesis of r… Show more

“…They always had both sister chromatids of each chromosome identically labelled. All the 150 labelled tetraploid (plate I) and 33 labelled octaploid plates showed the labelling pattern first described by Taylor et al (1957) and Taylor (1958). Chromosomes with label in one chromatid only and those with label in the non-homologous segments of the two sister chromatids (the latter brought about by exchange) were found more or less in equal numbers (48 and 52 per cent, respectively).…”

“…(v) Peacock (1963) made experiments on Vicia faba similar to those of Taylor et al (1957) and La Cour and Pelc (1958). He found that colchicine neither influenced replication nor the frequency of sister chromatid exchange.…”

“…There are numerous methods for measurement of cell proliferation, including quantification of cells that incorporated [ 3 H] thymidine or BrdU (13,14), CFSE labeling (15), or direct detection of recently divided cells by Ki-67 expression (16). Earlier studies using these methods suggested intrathymic expansion of mature thymocytes before export to the periphery (17).…”

“…^H-Tdr, which was introduced by Verly et (1958) and Taylor et (1957) as a specific label for DNA, has provided a means not only for the detailed analysis of cell cycle dura tions, but for kinetic studies of cell populations in animal tissues as well. With this aid it has been possible to label cells that are synthesizing DNA which permits study of their division and their subsequent growth, maturation, function and death with great precision.…”
