The ORP9-ORP11 dimer promotes sphingomyelin synthesis

Abstract: Numerous lipids are heterogeneously distributed among organelles. Most lipid trafficking between organelles is achieved by a group of lipid transfer proteins (LTPs) that carry lipids using their hydrophobic cavities. LTPs localize at membrane contact sites (MCS) where donor and acceptor organelles form a synapse to exchange information and material. The human genome encodes 90 intracellular LTPs responsible for lipid trafficking and the function of many LTPs in defining cellular lipid levels and distributions … Show more

“…Recently, metabolic incorporation of azide containing L-serine 5 (C-L-Ser-N 3 ) and myo -inositol 6 (Ino-N 3 ) have been shown to label phosphatidylserine (PS) and phosphatidylinositol (PI), respectively. Both these lipid classes are synthesized in the ER and have been proposed to be trafficked to the cytoplasmic leaflet of the trans-Golgi and TGN through lipid transfer proteins 31,32 . When analyzed with FACES, we observed 1a -PI in Golgi cisternae that was evenly distributed between luminal and cytoplasmic leaflets (Fig.…”
