The Palatability of Lopinavir and Ritonavir Delivered by an Innovative Freeze-Dried Fast-Dissolving Tablet Formulation

Abstract: Negative hedonic sensory qualities of HIV antiretroviral drugs often reduce patient adherence particularly in pediatric populations requiring oral consumption. This study examines the palatability of an innovative delivery mechanism utilizing a freeze-drying-in-blister approach to create fast-dissolving tablets (FDTs) containing a fixed-dose combination of lopinavir and ritonavir (LPV/r). Consumption patterns of solutions during brief-access and long-term testing and baby foodstuff consumption were analyzed to… Show more

“…Research shows that both palatability and swallowability of medications remain a challenge in children under four years of age [ 26 ] especially for Lopinavir/Ritonavir formulations where caregivers try to crush the pills to facilitate swallowing. Though this consequently reduces bioavailability and efficacy of the LPV/r [ 27 ].…”

“…Both caregivers and HCPs identified the appealing flavour as a clear advantage over the previous bitter-tasting formulations. 21 Adverse effects, such as nausea, taste disturbances or reduced appetite, have been reported with all ARVs and are the main reason for discontinuing, switching or non-adherence. 22 Initial immediate reactions of children towards the medicines by 'vomiting' in our study were overcome through practical support provided by the HCPs.…”
