The Penetration of Moxifloxacin into the Pancreas of Male Rats in Experimental Acute Necrotizing Pancreatitis

Abstract: Infectious complications of acute necrotizing pancreatitis (ANP) determine the extent of multiorgan failure and account for 80% of deaths. Prophylactic use of antibiotics can reduce the incidence of these complications. However, the actual indication as well as choice of drug remains a controversial matter. We examined the penetration of moxifloxacin, a new broad-spectrum fluoroquinolone, in healthy and inflamed pancreatic tissue in rats after inducing ANP. The concentration of moxifloxacin in pancreatic tissu… Show more

“…Pharmacokinetics of DCQD components magnolol, hesperidin and naringin were obviously lower in SAP group than in control group, suggesting that lack of an effective blood volume and a systematic inflammatory response to organ damage in SAP rats would affect the distribution, metabolism and excretion of DCQD components [15] . No significant difference was found in T1/2α and Tmax of DCQD components such as magnolol and naringin between the two groups, which may be due to the way of modeling experiments.…”

“…In agreement with the moxi assay [14], the HPLCinstrumentation consisted of a Shimadzu HPLC 10 A system, comprising a thermostated column compartment, a quaternary pump, a degasser, a system controller, an auto injector with sample cooler and spectrofluorometric detector. Data integration was performed using the CLASS LC 10 ® software (Shimadzu Europe GmbH, Duisburg, Germany).…”

“…In this report, a previously published HPLC assay for the determination of moxifloxacin (moxi) in serum and pancreas tissue [14], which used ofloxacin as internal standard, was adapted to allow the simple and rapid determination of levo in plasma and dialysate, using moxi as internal standard. Since the assay was also aimed to serve for pharmacokinetic studies of levo during continuous haemodialysis measurement of the drug in dialysate was necessary.…”

“…Conventional preparation of blood or plasma samples and quantitative determination by HPLC of linezolid, daptomycin, and moxifloxacin was achieved as published previously [18][19][20] using Shimadzu (Kyoto, Japan) series 10A instruments. Samples of PBS were subjected to HPLC without pre-treatment.…”
