The Phase Diagram of Charged Colloidal Lipid A-Diphosphate Dispersions

Abstract: Small-angle X-ray-scattering, light-scattering, and electron microscope experiments were used to determine the phase transitions of colloidal lipid A-diphosphate aqueous dispersions. The phases detected were a correlated liquid phase, a face-centered cubic (Fd3m) and a body-centered cubic (Im3m) colloidal crystal phase and a new glass phase. These experimentally determined phases were shown to be in accord with theoretically predicted equilibrium phases.

“…This was shown by Revier and Aste 55 for tetrahedrally close-packed ͑tcp͒ ensembles, as well and in a study of lipid A-diphosphate. 56 For the structure reported by Revier and Aste, 55 face and edge angles were close to the ideal values of 120°and 109.47°. In the Pm3n and Fd3m cellular networks there are two examples of 24 tcp structures.…”

“…35,56 This would require the presence of diffusive planes in reciprocal space, however, this was not observed. The transverse extension, l −1 , of the diffusive planes was smaller than the resolution; hence under the given con- ditions it was very small compared to 1 / a.…”

“…The phase transitions were a correlated liquid phase, a cubic FCC and a body-centered cubic crystalline phase [35]. These phases were detected in the presence of mM NaCl for different volume fractions, ; and various crystal forms (BCC and FCC) could be obtained.…”

“…However, a mixture of equimolar concentrations of the two antagonistic molecules revealed a SAXS-powder diffraction pattern a light scattering profile for crystals of sizes of 1 m that could be indexed for a much larger face-centered (Fd3m) unit cell, with a = 58.0 nm. However, when employing the similar conditions for colloidal Lipid A-diphosphate dispersions but through addition of M NaOH rather than by removing NaCl [35] a very different phase behavior was observed (Fig. 2).…”
