The phosphoinositide dependent-phospholipase C pathway differentially controls the basal expression ofDREB1andDREB2genes

Abstract: W e recently showed that-in Arabidopsis thaliana suspension cells-phosphoinositide dependent-phospholipase C (PI-PLC) and diacylglycerol kinase (DGK) negatively regulated the basal expression of most DREB2 genes. DREB2 genes encode transcription factors that bind to Drought Responsive Elements (DRE). Those elements are also bound by DREB1 factors. While DREB2 factors are mostly involved in drought and heat responses, DREB1s are induced in the response to chilling. We here show that the pharmacological inhibiti… Show more

“…Glycerophospholipids like phosphatidic acid (PA) (FC = 17.72), phosphatidylcholine (FC = 6.56), phosphatidylinositol (FC = 5.77) are known to be deposited to enforce cell walls. Furthermore, these are also known to perceive and transmit signals activating downstream genes eventually regulating R genes to biosynthesize RR metabolites and RR proteins [ 43 ]. These compounds are either converted into bioactive lipids (components of lipid bilayer of cell membrane) or stay as soluble molecules (messengers), further binding to the downstream mitogen activated protein kinases (MAPK), further affecting the enzymatic activities, vesicle trafficking and ion fluxes [ 43 ].…”

“…There is a special and thoroughly studied subgroup, IIIc, namely the DREB1s. The AtDREB1 factors are mostly induced in response to chilling [ 88 ]. Similar molecular mechanisms and physiological responses, induced and regulated by DREB1 genes, appear to occur in monocots [ 5 , 25 , 65 , 89 ].…”

“…Some subgroup IIId genes may have constitutive expression or participate in responses to abiotic stress, for example, OsDREB4-2 was induced by drought and salt stresses, but OsDREB4-1 was constitutively expressed [ 90 ]. The group IV genes were designated as DREB2 genes, although they were reported as mostly involved in drought and heat responses [ 88 ]; however, they may have more functions, such as DREB2A being involved in drought-, salt- and heat-stress responses in Arabidopsis [ 91 ]. AtDREB2C -overexpressing Arabidopsis plants were dehydration sensitive, and were freezing and heat tolerant [ 92 ].…”

“…Two of these, phospholipid‐remodeling into galactolipids through the generation of phosphatidic acid (PA) by phospholipase C (first route) or phospholipase D (second route), have been extensively studied in connection to phosphate deficiency responses (Gaude, Nakamura, Scheible, Ohta, & Dörmann, ; Nakamura et al., ). We do not find either of them to respond to increased irradiance, although they are known to repress expression of the transcription factor DREB2A (Figure ; (Ruelland, Djafi, & Zachowski, )). The activation of the GDPD pathway (third route), as a result of Pi limitation, probably outcompetes the PLC pathway, leading to reduced PLC‐mediated repression of DREB2A .…”
