The photocycle of the chloride pump halorhodopsin. II: Quantum yields and a kinetic model

Abstract: The quantum yield of the primary photoreaction of HR578 was determined as øgreen = 0.34±0.02 and that of the photochemical reconversion of HRL410 into HR578 as øblue = 0.01±0.004. From steady‐state illumination and flash‐photolysis experiments, a quantitative description of HRL410 formation and decay was made leading to a model of the photocycle in isolated, light‐adapted HR. This model satisfies all known facts about HR including its photochromic behaviour.

“…This result was confirmed by measuring the yield of the primary reaction of K formation under various conditions [14]. A value of >0.6 was determined again for M formation in basal salt/ether [6] and this value was confirmed by Kouyama et al [15] using the same system. Using resonance Raman scattering experiments as an alternative method to measure the quantum yield a value of 0.67 [16] was determined.…”

“…This value was redetermined a couple of years later with a different experimental setup [6]; due to the light scattering a lower limit of 0.6 was found. Several research groups determined the quantum efficiency for the step BR , K at -196°C as 0.28 [7], 0.5 [8] and 0.33 [9].…”

“…D137 (Hhbop D96), which is involved in the reprotonation of the Schiff's base from the cytoplasmic side, is found only in BRs. D126 (Hhbop D85), which is involved in the deprotonation of the Schiff's base to the external side, is not found in HRs, and, correspondingly, no deprotonated intermediate is found in the photocycle of HR of H. halobium (22,34 (30,44,45).…”

“…The first of these tools was the chloride pump halorhodopsin derived from the halobacterium Nathomonas pharaonis (NpHR) Oesterhelt et al, 1985). NpHR when activated by green/yellow light (peak absorbance *570 nm) moves chloride ions inside the cell, hyperpolarizing the neurons, and inhibiting spiking.…”
