The polar lipid composition of walsby's square bacterium

Abstract: Square, gas vacuole-containing Archaea of the type first described by Walsby were found to dominate in a saltern crystallizer pond in Eilat, Israel. To obtain information on the taxonomic position of these yet uncultured bacteria, we analyzed the polar lipids present in the microbial community in the saltern brine. In addition to phosphatidylglycerol, phosphatidylglycerophosphate and phosphatidylglycerosulfate we found one glycolipid, chromatographically identical with the sulfated diglycosyl diether lipid fou… Show more

“…Cellular fatty acids of photoheterotrophically grown cultures were analysed as described by Sasser (1990). Polar lipid analyses were performed using 1 g freeze-dried cells of photoheterotrophically grown cultures as described previously (Subhash et al, 2013b, c;Oren et al, 1996). Quinone analysis was performed after extraction with chloroform and methanol (2 : 1, v/v) by previously described methods (Imhoff, 1984;.…”

“…Polar lipid extraction and characterization by TLC on silica gel plates (20i20 cm ; Sigma) was performed as described by Torreblanca et al (1986), using single development in chloroform\methanol\acetic acid\water (85 : 22n5:10:4, v\v). Plates were stained for glycolipids and other lipids using the α-naphthol stain, the CeSO % \H # SO % stain, the orcinol\FeCl $ stain and the molybdate stain for detection of phospholipids (Kates, 1972 ;Oren et al, 1996 ;Torreblanca et al, 1986). The presence of C #!…”
