Production of mini-tubers as a source for seed potato was investigated by growing in soil micropropagated plants and micro-tubers produced from micropropagated plants . Cultures of several cultivars were initiated from indexed tubers and multiplied on modified MS medium . Cultures were micropropagated by using a modular system which allowed batch handling . Micropropagated plants produced mini-tubers in glasshouse after 70-115 days of growth in soil . A large proportion of the mini-tubers produced were between 9 and 15 mm diameter . Several factors, e.g ., explant number, duration of in vitro culture and genotype influenced mini-tubers production . Micropropagated plants after culture of 86 days or longer produced micro-tubers ca . 2 to 10 mm diameter. Plants, which formed micro-tubers in vitro, produced less number of mini-tubers in soil . Micro-tubers produced 1 to 3 mini-tubers when grown in soil in chain-type paper pots, but produced conventional sized tubers when grown in soil under plastic polytunnel . Mini-tuber number varied widely between potato cultivars ; cvs. Bintje and British Queen produced more mini-tubers than the other cultivars . Mini-tubers developed green hard skins when kept in light for 3 weeks, and could be stored in dark at 4° C upto 6 months . In a field trial, small mini-tubers ca . 5-10 mm diameter produced more but smaller tubers than mini-tubers ca . 15-20 mm diameter. The micropropagated plants and the plants grown from mini-tubers were genetically stable, and did not show any morphological aberrations except for one variegated plant among the several thousand produced . It is concluded that the production of mini-tubers by soil planting of micropropagated plants is a rapid and efficient method for producing seed potato tubers .

Drought stress is a major abiotic constraint limiting crop production worldwide. Screening for drought tolerance and the traits that enhance drought tolerance is not straightforward in large mapping populations. In this study, we investigated the possibility of screening a mapping population in vitro for PEG-induced water deficit stress and recovery potential. We have measured several shoot and root growth parameters or traits in the C 9 E diploid potato mapping population. Significant variation was observed for genotype-specific responses to water deficit and recovery potential. Genetic variation and heritability estimates were high to very high for the measured traits depending on growth conditions. In order to identify potato QTLs for drought tolerance and recovery potential an SNP marker-rich integrated linkage map was used. A total of 23 QTLs were detected under control, stress and recovery treatments explaining 10.3-22.4% of the variance for each phenotypic trait. Among these, 10 QTLs were located on chromosome 2. Three QTLs involved in the important trait root to shoot ratio were identified on linkage groups 2, 3 and 8. These loci explained together 41.1% of the variance for this trait, and may be breeding targets for stress tolerance and yield in the field as well. The SNP markers derived from EST sequences underlying these QTLs led to the identification of putative candidate genes for further study in potato. This study constitutes the first knowledge of in vitro screening of a mapping population for drought tolerance in potato.

With the objective to develop a practical method of screening potato for drought tolerance, shoot and root growth in plantlets raised in vitro (from nodal cuttings drawn from in vivo as well as in vitro grown plantlets) were studied in three genotypes with known root mass production under field conditions. Different levels of water stress were induced using five concentrations of agar in MS (Murashige and Skoog in Physiol Plant 15:473-497, 1962) medium. Water potential of various media ranged from −0.70 MPa to −0.98 MPa. Water stress in culture adversely affected plantlet growth, and the responses varied with genotype and explant source. Genotype IWA-1 was less affected than Konafubuki and Norin-1. In the experiment with explants from in vivo grown plants, the time to rooting was considerably delayed in Konafubuki and Norin-1 by an increase in agar concentration, but no such effect was observed in IWA-1. In all media, the mean number of roots and root length was greater in IWA-1 than Konafubuki and Norin-1, and the latter two genotypes were at par. At 10 gl −1 agar, IWA-1 had taller plantlets, heavier foliage dry weight, root volume, as well as root dry weight than Konafubuki and Norin-1, whereas the latter two genotypes were at par for all these characteristics. This pattern was similar to the reported pattern of these genotypes for root dry weight under field conditions. However, such similarity in the in vitro and field behavior of the tested genotypes was not observed when nodal cuttings drawn from in vitro plantlets were used as explants. It is concluded that in vitro screening of potato under specific and limited water stress conditions by raising plantlets from nodal cuttings drawn from in vivo grown plants may provide a system for effectively differentiating the genotypes for their expected root mass production under field conditions.
