The potential roles of retinoids in combating drug resistance in cancer: implications of ATP-binding cassette (ABC) transporters

Abstract: Multidrug resistance (MDR) means that tumour cells become unresponsive during or after the course of treatment to one or more of chemotherapeutic drugs. Chemotherapeutic resistance critically limits the treatment outcomes and remains a key challenge for clinicians. The alternation in intracellular drug concentration through the modulation of its transport across the plasma membrane is the major cause for MDR and is adopted by various mediators, including ATP-requiring enzymes (ATPases). Among these ATPases, AB… Show more

“…Despite this, the development of drug resistance has proved to be a major obstacle to successful patient outcomes ( Wang et al, 2022c ; Karami Fath et al, 2022 ; Pastwińska et al, 2022 ). Chemotherapy exerts its cytotoxic effects by inhibiting cellular synthesis of DNA and RNA, suppressing cell proliferation, and promoting apoptosis ( Abdelaal and Haffez, 2022 ; Yang et al, 2022b ; Li et al, 2022d ). However, the efficacy of chemotherapy is limited by drug resistance, which leads to tumor progression and ultimately patient mortality.…”

“…Over the past 30 years, there have been three generations of P-gp modulators developed as a result of research on the reversal of MDR; due to a variety of reasons, however, these modulators failed to show effectiveness in clinical trials. Part of the reason for the failure was that the medication lacked bioavailability at tumor sites and caused nonspecific toxicities (Abdelaal and Haffez 2022 ).…”

“…Cancer is an important factor threatening human health, with high mortality and recurrence rates. At present, the results of cancer treatment have been unsatisfactory, especially single chemotherapeutic drugs showing non-selective side effects and multidrug resistance. Therefore, developing an effective multi-mode combined therapy platform plays an important role in tumor treatment. , Photodynamic therapy (PDT), among others, has drawn a lot of interest as a non-invasive approach to treating cancers due to its exceptional spatial and temporal precision and synergistic therapeutic advantages. , Under appropriate wavelength of laser, the reactive oxygen species (ROS) produced by photo-sensitizers react with biomacromolecules (cellular phospholipids, nucleic acids, proteins, etc. ), leading to cell damage and necrosis. , Common photosensitizers mainly include hematoporphyrin, modified porphyrins, dihydromorphines, phthalocyanines, and natural macromolecules .…”
