The Pulmonary Neuroendocrine System: The Past Decade.

Abstract: The pulmonary neuroendocrine system consists of specialized airway endocrine epithelial cells, associated with nerve fibres. The epithelial cells, the pulmonary neuroendocrine cells (PNEC), can be solitary or clustered to form neuroepithelial bodies (NEB). During the last thirty years, the pulmonary neuroendocrine system has been intensively investigated and much knowledge of its function has been obtained. This text reviews work which dates from the last ten years. In this period, the picture of the pulmonary… Show more

“…However, we cannot rule out similar regulatory functions for other PNEC-derived neuropeptides. PNECs have been implicated as regulators of ventilationperfusion mismatches in response to hypoxia (16,58). Bronchoconstrictor and vasoconstrictor responses could be homeostatic in normal lung, maintaining optimal ventilation-perfusion matching despite regional hypoxia.…”

“…In contrast, hyperplastic and embryonic NEBs accumulate PAMP and PAM, but not AM (Table 1). These results clearly support the earlier suggestions (Van Lommel et al, 1999) that PNECs and NEBs are biologically different entities with distinct physiological properties and probably different functional roles.…”

“…A variety of peptides with several proposed biological functions, such as bombesin and CGRP, are produced and secreted by pulmonary NE cells (Van Lommel et al, 1999). Several studies have provided strong support for the relationship between pulmonary NE cells and intrauterine and postnatal pulmonary growth and maturation (Emanuel et al, 1999;Sorokin et al, 1997).…”

“…NECs are present within the airway epithelium of the lung, often adjacent to small pulmonary vessels. 36 They synthesize and secrete a variety of neuropeptides and amines, including BLPs, ET-1, 5-HT, and likely sub P. 37,38 Hyperplasia of lung NECs has been described in association with exposure to injurious stimuli, leading to structural remodeling of the PA wall and PHTN. 37,39 -44 In addition, hyperplasia of NECs has been detected in response to NEP inhibition.…”
