The Quantitative Assay of Corticotrophin Using Rats Treated With Hydrocortisone Acetate

Abstract: An attempt has been made to modify the Sayers method for the assay of corticotrophin by substituting for hypophysectomy an injection of sufficient hydrocortisone acetate to suppress the release of endogenous pituitary corticotrophin in intact rats over the period of the assay. The results of forty such assays of subcutaneous corticotrophin seem to indicate that this procedure is about 1+ to 2 times as efficient as the original method using hypophysectomized rats. For a 2 + 2 point assay it was necessary to use… Show more

“…Biological assays 'Total gonadotrophin' was assayed by the uterine weight method, FSH by the ovarian augmentation method (Butt et al 1961) and luteinizing hormone (LH) by the ovarian ascorbic acid depletion method (Parlow, 1958(Parlow, , 1961. In the latter the ascorbic acid in one ovary from each rat was measured by the dichlorophenolindophenol method (Dekanski & Harvie, 1960) and the gonadotrophin injected intra¬ peritoneally in a volume of 0-2 ml. saline or buffer.…”

“…The adrenal glands were quickly removed from the dorsal, weighed and homogenized in metaphosphoric acid. The determination of the ascorbic acid concentration was carried out with the dichlorphenol-indophenol method, described by Dekanski and Harvie (1960). The values obtained were calculated in jug to 100 mg adrenal wet weight.…”
