The Regulation of the Proliferation and Differentiation of Rat Leydig Cell Precursor Cells After EDS Administration or Daily HCG Treatment

Abstract: The proliferation and differentiation of possible Leydig cell precursors in adult rats were studied after destruction of the existing Leydig cells with EDS or after daily treatment with hCG. After 2 days with either treatment, a 12- to 16-fold increase in the number of [3H]thymidine-incorporating interstitial cells was found. In the case of hCG treatment, this was probably due to the high plasma hCG levels. However, after EDS treatment, LH levels start to rise between days 1 and 3, suggesting a paracrine stimu… Show more

“…However, there is evidence that although the Leydig cell number is relatively stable, there is some turnover. For example, 3 H-thymidine has been shown to be incorporated into cells in the interstitial compartment (27), and injections of rats with high doses of hCG to result in the formation of new Leydig cells (28). These early studies, based on 3 H-thymidine labeling, in fact may have underestimated the turnover rate of adult Leydig cells.…”

“…EDS was administered in a single dose of 75 mg͞kg body weight. This treatment resulted in a rapid elimination of the LCs present in the adult testis by 4 days after treatment (46). Approximately 1,000,000 SLCs in a 25-l volume of PBS were injected into the parenchyma of recipient testes 4 days after the rats had received EDS.…”

“…Cell counts were carried out [16]. Briefly, nuclei of Leydig cells were counted; these were recognized by their more or less spherical nucleus with a typical distribution of heterochromatin and blue-purple-staining cytoplasm.…”

“…Peritubular cells and perivascular cells may be the stem cells or precursor cells of the Leydig cells [2,8,9,15,161. This assumption is supported by the fact that, after the destruction of the existing population of Leydig cells by EDS or daily hCG treatment (when the proliferative activity of intersti-tial cells is greatly enhanced), most labeled interstitial cells are located peritubularly and perivascularly [16]. Hence it cannot be excluded that in the normal adult rat new Leydig cells are formed not only as a result of proliferation of existing Leydig cells but from differentiation of Leydig cell precursors.…”
