The Renin–Angiotensin System Mediates EGF Receptor–Vitamin D Receptor Cross-Talk in Colitis-Associated Colon Cancer

Abstract: Purpose We previously showed that epidermal growth factor receptor (EGFR) promotes tumorigenesis in the azoxymethane/dextran sulfate sodium (AOM/DSS) model, whereas vitamin D (VD) suppresses tumorigenesis. EGFR-vitamin D receptor (VDR) interactions, however, are incompletely understood. VD inhibits the renin-angiotensin system (RAS), whereas RAS can activate EGFR. We aimed to elucidate EGFR-VDR cross-talk in colorectal carcinogenesis. Experimental Design To examine VDR-RAS interactions, we treated Vdr+/+ and… Show more

“…RAS is mitogenic and angiogenic, and contributes to neoplastic growth in ovarian, prostate, lung, breast and pancreatic cancers (25). A number of RAS components, including renin, renin-receptor, ACE and AngII, are locally upregulated in tumors (26). In the present study, these molecules were significantly reduced by elemene treatment, suggesting that its antitumor effects may involve downregulation of the RAS signaling pathway.…”

“…Ang II stimulates their proliferation and induces production of various cytokines, such as TGF‐β, some of which have immunosuppressive functions. CAF, which are considered to be immunosuppressive cells in the cancer microenvironment, also express AT1R . Thus, we evaluated the effect of ARB treatment on CAF phenotypes in our mouse models.…”

“…CAF, which are considered to be immunosuppressive cells in the cancer microenvironment, 4 also express AT1R. 27 Thus, we evaluated the effect of ARB treatment on CAF phenotypes in our mouse models. For sorting of pure CAF, we used specially devised tumor-bearing mouse models in which MC38 cells labeled by a fluorescent protein, DsRed, were inoculated into CAG-EGFP transgenic mice that expressed EGFP ubiquitously.…”

“…No study has focused on the effect of vitamin D3 supplementation on ACE expression. However, it has been observed that VDR knockout mice display an increased production of ACE mRNA [ 68 ]. Interestingly, this metalloprotease is involved in Alzheimer’s disease since human ACE increases degradation and cleavage of Aβ [ 69 72 ].…”
