The role of bacterial DNA containing CpG motifs in diseases

Abstract: Bacterial DNA containing unmethylated CpG motifs can activate immune cells to release proinflammatory cytokines. Here, the role of bacterial DNA containing CpG motifs in diseases with a focus on arthritis is discussed. Our studies demonstrate that the intraarticular injection of bacterial DNA and oligodeoxynucleotides containing CpG motifs (CpG ODN) induced arthritis. The induction of arthritis involves the role of macrophages over other cells such as neutrophils, NK cells, and lymphocytes. TNF-and TNFRI play … Show more

“…In addition, the retention of bacterial DNA in the formulated proteoliposomes also favors lymphocyte activation through the specific nucleotide binding to Toll-like receptor (TLR) 9 in APCs. Demethylated CpG DNA induces the expression and secretion of IL-6, TNF-α and IL-12, promoting the immune response [ 59 , 60 ].…”

“…Moreover, our former findings reported that the sustained upregulation of IFN-γ was advantageous to bacterial clearance [ 11 , 23 ]. Furthermore, in animals, IL-12 can activate the natural killer cells to produce IFN-γ, while in return, IFN-γ can activate macrophages to produce IL-12, so a self-amplifying loop is formed [ 58 , 59 ]. The higher expression of IL-12 in this study indicated that LACpG10-HL could boost the loop to induce Th1 immune responses.…”

“…CpG DNA is a type of DNA sequence with immune activation function containing unmethylated CpG motif, including artificially synthesized oligodeoxynucleotides containing CpG (CpG ODN) and genomic DNA of bacteria, viruses, and invertebrates (Zhou and Deng, 2021). CpG DNA triggers immunostimulatory activity through TLR9 (Ohto et al, 2015).…”
