The role of cartilage glycosaminoglycan structure in gagCEST

Abstract: Glycosaminoglycan (GAG) chemical exchange saturation transfer (gagCEST) is a potential method for cartilage quality assessment. The aim of this study was to investigate how the gagCEST effect depends on the types and molecular organization of GAG typically found in articular cartilage. gagCEST was performed on different concentrations of GAG in various forms: free chains of chondroitin sulfate (CS) of different types (-A and -C) and GAG bound to protein in aggregated and nonaggregated aggrecan extracted from c… Show more

“…These effects could result in a signal loss of 5% (based upon a T2 relaxation time in osteoarthritic tibial plateaus of 29.1 ms 16 and the chosen TE of 1.4 ms). Other confounding effects could be the GAG type in human cartilage, which has been shown in recent work by Einarsson et al 17 to be of influence in gagCEST measurements.…”

“…The results of qPCR analysis showed that gene expression levels of chondroblasts did not increase significantly within 7 to 14 days. ACAN expression and extracellular matrix play an important role in the differentiation and maturation of chondroblasts 31 . Sox9 plays a key role in chondrocyte differentiation, and its expression is associated with multiple stages of chondrocyte differentiation 32 .…”

“…CEST signal studies of different forms of GAGs typically found in articular cartilage recently demonstrated that the CEST effect arises mainly from chondroitin 4-sulfate. 43 Swarm rat CHS was demonstrated to highly express aggrecan, which is composed of a core protein with covalently attached side chains of linear GAGs, including a high number of chondroitin sulfate, especially chondroitin 4-sulfate. 44 The GAG CEST approach was combined with 99m Tc-NTP 15-5 nuclear imaging, a radiotracer designed to target negatively charged molecules of GAGs.…”
