The Role of Clonal Hematopoiesis of Indeterminant Potential and DNA (Cytosine-5)-Methyltransferase Dysregulation in Pulmonary Arterial Hypertension and Other Cardiovascular Diseases

Isaac M. Emon,

et al.

Abstract: DNA methylation is an epigenetic mechanism that regulates gene expression without altering gene sequences in health and disease. DNA methyltransferases (DNMTs) are enzymes responsible for DNA methylation, and their dysregulation is both a pathogenic mechanism of disease and a therapeutic target. DNMTs change gene expression by methylating CpG islands within exonic and intergenic DNA regions, which typically reduces gene transcription. Initially, mutations in the DNMT genes and pathologic DNMT protein expressio… Show more

“…DNA methylation is an example of an epigenetic mechanism. DNA methylation is facilitated by enzymes called DNA methyltransferases (Dnmt1 (DNA cytosine-5 methyltransferase 1; EC 2.1.1.37), Dnmt3a (DNA cytosine-5 methyltransferase 3α; EC 2.1.1.37), and Dnmt3b (DNA cytosine-5 methyltransferase 3β; EC 2.1.1.37)) [40,41]. Rats that received prolonged IH exhibited an increased Dnmt enzyme activity and high levels of Dnmt 1, 3a, and 3b proteins, which corresponded to changes in their respective mRNA levels [36][37][38].…”
