The role of dopamine D3, 5-HT2A and 5-HT2C receptor variants as pharmacogenetic determinants in tardive dyskinesia in African-Caribbean patients under chronic antipsychotic treatment

Abstract: Tardive dyskinesia (TD) is associated with polymorphisms of the dopamine D(3), serotonin 2A and 2C receptors (DRD3, HTR2A and HTR2C, respectively). This study investigated the possible relationship between TD and the polymorphisms Ser9Gly (DRD3), 102T>C (HTR2A), -1438G>A(HTR2A) and Cys23Ser (HTR2C) in African-Caribbean inpatients. One hundred and twenty-six patients under chronic antipsychotic treatment were genotyped. The assessment of TD was carried out with the abnormal involuntary movement scale (AIMS). Th… Show more

“…A meta-analysis found an association of the C allele in the 102T/C polymorphism of HTR2A with TD. 164 Nonetheless, there are numerous negative findings for this SNP as well (for example, see Wilffert et al 165 ). Studies also reported positive results with the -1438G/A 166 in HTR2A and the Cys23Ser polymorphism in HTR2C (for example, see Al Hadithy et al 156 ).…”

“…Of these, 18 assessed the association between TD and DRD3 Ser9Gly. Two were excluded because the TD outcome was only examined continuously using mean AIMS scores [Basile et al, 1999;Wilffert et al, 2008]. Two conference abstracts were also excluded [Utsunomiya et al, 2006;Zai et al, 2007a].…”

“…Cardiovascular ranked second with Puerto Rico [25][26][27][62][63][64] and Costa Rica [11,15,16,50,60,65,113] as the main contributors. Neurology/psychiatry placed third with a greater contribution of the Caribbean islands of Curaçao [85,86,110], Martinique [87], Jamaica [56,61], Cuba [82] and Puerto Rico [35,36,66,95] (Figure 3). These results are in agreement with the global trend of scientific research in these areas [10].…”
