The role of endothelial leak in pulmonary hypertension (2017 Grover Conference Series)

Abstract: The canonical transient receptor potential 4 (TRPC4) protein contributes to the molecular make-up of endothelial store-operated calcium entry channels. Store-operated calcium entry is a prominent mode of calcium influx in endothelium. Store-operated calcium entry channels are activated by inflammatory mediators and growth factors, and in endothelium, this process induces inter-endothelial cell gaps that increase permeability. Pulmonary endothelium within extra-alveolar segments, including pulmonary arteries, i… Show more

“…We were more likely to obtain viable PAECs from PAH patients with more severe disease (as evidenced by hemodynamics and possibly shorter 6MWD), which in part may be related to our observation that PAH PAECS were more apoptosis resistant compared to commercial PAECs. Altered mechanical properties (i.e., hyperpermeability) of PAH PAECs may have lead to increased transfer from the endothelium to the PAC balloon, as has been shown in experimental PH (13). These observations are preliminary and require confirmation.…”

“…The role of TRPC4 in PAH was investigated in a model of hypoxic PAH in which rats were pretreated with a vascular endothelial growth factor receptor 2 inhibitor, exposed to three weeks of hypoxia, and then returned to normoxia. In this model, TRPC4 contributed to the hyper-permeability of endothelial cells, demonstrating a novel type of endothelial abnormality in severe experimental PAH [ 168 ].…”
