The Role of Exosomal Epigenetic Modifiers in Cell Communication and Fertility of Dairy Cows

Abstract: Abnormal uterine function affects conception rate and embryo development, thereby leading to poor fertility and reproduction failure. Exosomes are a nanosized subclass of extracellular vesicles (EV) that have important functions as intercellular communicators. They contain and carry transferable bioactive substances including micro RNA (miRNA) for target cells. Elements of the cargo can provide epigenetic modifications of the recipient cells and may have crucial roles in mechanisms of reproduction. The dairy i… Show more

“…UC followed by SEC of plasma samples may therefore contain uniquely sorted exosomal cargo and provides a better crosssection of genetic materials including miRNA. Differential miRNA expression in EX and non-EX samples suggest the involvement of miRNA in the regulation of many different biological functions at both the cellular and systemic level (Zhang et al, 2019;Abeysinghe et al, 2020). Highly expressed miRNAs from maternal EV populations relates to key bovine traits of interests.…”

“…EVs are classified into subpopulations by their origin, size, morphology, and protein markers specific to each subtype. Exosomes (EX) are an EV subtype of diameter ~30 -150 nm and carry unique molecular cargo that has been used in biomarker development and targeted therapeutics (Zhang et al, 2019;Abeysinghe et al, 2020;Dai et al, 2020;Kugeratski et al, 2021). Exosomal cargo contains complex functional molecules ranging from proteins (Crookenden et al, 2016), lipids (Record et al, 2014), mRNAs (Valadi et al, 2007) and miRNAs (Valadi et al, 2007).…”

“…Epigenetic regulation of gene expression has been well studied with regards to mammalian development [ 70 , 71 , 72 , 73 , 74 ]. However, a new area of epigenetics is developing following research into the role of miRNAs as epigenetic modulators and has been reviewed recently [ 75 , 76 ]. Briefly, the epigenome is controlled at the base level by the expression of genes that encode for a group of enzymes, termed DNA methyltransferases (DNMTs) [ 71 , 73 ].…”
