The role of fungal tremorgens in ryegrass staggers

Abstract: Administration of the fungal tremorgens penitrem A, verruculogen, fumitremorgin B, and paxilline to sheep produced effects similar to signs of ryegrass staggers (RGS); fungi capable of producing tremorgens were present in the environment where RGS was occurring or had occurred; and spores of these fungi were ingested by sheep grazing the pastures. Of 23 species of Penicillium isolated from toxic pastures, 2 common species produced tremorgens: P. crustosum produced penitrem A and P. piscarium produced either ve… Show more

“…7 x 10-7M) and a 150% increase in Vmax (Vmax = 5.0 x 10-13 mole/,ug P/min) relative to control values (Km = 2.2 x 10-7M; Vmax = 2.0 x 10-l3 mole/4g P/min). These values returned to approximate control levels by1 week post-injection (Km = 2.6 x 10-7M; Vmax = 2.13 mole/tg P/min), and then decreased by 77% (Km = 0.5 x 10-M) and 45% Vmax = 1.1 x 10-'3 mole/pg P/min) by 2 weeks. Statistical analysis of these data Lineweaver-Burk plot of 3H-glutamate uptake.…”

“…Uptake: In Vivo Toxin Exposure. Rats (N = 12) were given intraperitoneal injections of either DMSO or a tremorgenic dose of aflatrem (3 mg/kg IP), and decapitated at 24 hr, 1 week, and 2 weeks after the injection. Day of injection was staggered so that any given experiment contained tissue from rats killed at each time point post-injection.…”

“…Molds producing such tremorgens pose a recognized health risk to humans and animals when these microorganisms are present in spoiled food or feed. For example, livestock grazing on moldy pastures may develop sporadic locomotor incoordination, where they may stagger, fall over, and suffer muscular spasms [5], [6]. However, the mode of action for many of these mycotoxins has not been elucidated in detail.…”
