The role of microRNAs in bone remodeling

Abstract: Bone remodeling is balanced by bone formation and bone resorption as well as by alterations in the quantities and functions of seed cells, leading to either the maintenance or deterioration of bone status. The existing evidence indicates that microRNAs (miRNAs), known as a family of short non-coding RNAs, are the key post-transcriptional repressors of gene expression, and growing numbers of novel miRNAs have been verified to play vital roles in the regulation of osteogenesis, osteoclastogenesis, and adipogenes… Show more

“…They can induce changes in multiple cellular processes, including cell proliferation, cell differentiation, and cell senescence (Davis et al., 2017; Jing et al., 2015). However, their instability in the extracellular environment limits their application.…”

“…MicroRNAs (miRNAs), which function as the key post‐transcriptional repressors of gene expression in diverse cells, are a class of short noncoding RNAs (~22 nucleotides; Jing et al., 2015; Li, Cheng et al., 2015; Wang et al., 2013). They can inhibit target gene levels by binding to the 3′‐untranslated region (UTR) of specific mRNA targets and hence degrade the mRNA or suppress translation.…”

“…These results together with the increasing knowledge, that miRNAs do impact on key regulators of osteoblast and osteoclast biology [2,3] prompted us to look more broadly at miRNAs differentially circulating in recent osteoporotic fracture patients [10], as well as in prevalent osteoporotic fractures, where, indeed, we identifi ed a signature of 4 miRNAs with predictive power superior to the gold standard of DXA measurements (Heilmeier et al in revision). In order to test, if these miRNAs also play a role in bone biology, we tested them for modulation of osteogenic differentiation, which several of these did indeed infl uence.…”
