The role of O‐linked sugars in determining the very low density lipoprotein receptor stability or release from the cell

Abstract: The very low density lipoprotein receptor is a member of the low density lipoprotein receptor supergene family for which two isoforms have been reported, one lacking and the other containing an O-linked sugar domain. In order to gain insight into their functionality, transient and stable transformants separately overexpressing previously cloned bovine variants were analyzed. We report evidence that the variant lacking the Olinked sugar domain presented a rapid cleavage from the cell and that a large amino-term… Show more

“…As previously reported for bovine aortic endothelial cells (60) and mammary carcinoma cells (61,62), VLDL-R-transfected HEK 293 cells expressed primarily the ϳ105-kDa isoform of the receptor lacking exon 16, which binds ligands identically to the larger ϳ130-kDa isoform (2,62).…”

“…N-terminal HCMV gN glycosylation has been suggested to protect from neutralization by antibodies (91). Moreover, dense glycosylation was found in the stem region of several glycoproteins in VZV, HCMV, and EBV, suspected to protect the otherwise vulnerable region from unspecific proteolytic cleavage (113). Another potential function could be to provide structural support for keeping the ectodomain away from the membrane (114).…”

“…The function of this Olinked sugar domain is still unclear. It may be responsible for controlling receptor recycling and degradation (31,32). In the present study, Northern analysis showed an apparent single transcript.…”
