The roles of PIKE in tumorigenesis

Abstract: Tumorigenesis is the process by which normal cells evolve the capacity to evade and overcome the constraints usually placed upon their growth and survival. To ensure the integrity of organs and tissues, the balance of cell proliferation and cell death is tightly maintained. The proteins controlling this balance are either considered oncogenes, which promote tumorigenesis, or tumor suppressors, which prevent tumorigenesis. Phosphoinositide 3-kinase enhancer (PIKE) is a family of GTP-binding proteins that posses… Show more

“…As we know, AGAP2 (ArfGAP With GTPase Domain, Ankyrin Repeat And PH Domain 2), also known as PIKE, belongs to the centaurin GRPase family, including three members, PIKE-L, PIKE-S, and PIKE-A [ 112 114 ]. PIKE-S and PIKE-L enhance PI3K antiapoptosis activity, but PIKE-A promotes the downstream Akt instead [ 113 , 115 ]. PIKE proteins participate in regulating the signal transducer and activator of transcription 5A (STAT5) by Janus kinase 2 (JAK2) phosphorylation [ 116 ].…”

“…Centg1 encodes phosphoinositide-3-kinase enhancer (PIKE) protein and GTPase family. PIKE is known as a proto-oncogene in tumor progression due to its anti-apoptotic activity [ 53 ]. PIKE is upregulated in a number of human cancer tissues including the colon and enhances cell transformation by Akt activation [ 54 ].…”

“…PIKE-L seems to be brain-specific 12 and its tissue-restriction distribution could be achieved by hypermethylation of the CpG island located on its promoter/first exon region (mapped as part of the ENCODE project). However, AGAP2 expression is considered ubiquitous and, furthermore, its levels are increased in several cancers (prostate cancer, glioblastoma and other tumours) and are associated to tumour progression 13 .…”
