The significance of alpha‐fetoprotein and other tumour markers in differential immunocytochemistry of primary liver tumours

Abstract: One hundred benign and malignant primary liver tumours were screened immunocytochemically for alpha-fetoprotein (AFP), alpha 1-antitrypsin, alpha-human chorionic gonadotropin, carcinoembryonic antigen (CEA), keratin and vimentin. Alpha-fetoprotein was found in 16/63 (24%) hepatocellular carcinomas and in two hepatoblastomas. When comparing tissue positivity for AFP with tumour differentiation, grade 1 hepatocellular carcinomas were found to be negative, while 21% of grade 2, 36% of grade 3 and 16% of grade 4, … Show more

“…Vimentin and bile duct-type cytokeratin expressions were scattered in HCC components. These findings in primary hepatic tumors may be interpreted in the context of their immaturity [2]. Such phenotypic variations of tumor cells in the present case may represent the stem cell nature of the neoplastic cells.…”

“…Positive immunostaining for AFP has been reported to be present in 2 to 61% of HCC (18). Expression of AFP can be focal and weak, leading to poor sensitivity, especially in small biopsies (2,7). Expression of AFP may be related to tumor grade, with higher grade HCC expressing AFP more frequently than lower grade tumors.…”

“…Immunohistochemical panels have been used to separate these tumors with variable success. ␣-Fetoprotein (AFP); polyclonal carcinoembryonic antigen (pCEA); and cytokeratins (CK) 7,8,18,19, and 20 are among some of the more commonly used markers in these panels (1)(2)(3)(4)(5)(6)(7)(8).…”

“…To seek the possibility to use GPC3 immunostaining to detect HCC component (cp) in CHC, combination of antibodies against GPC3, AFP, HepPar1, CK7 and CK17 were used. In addition to AFP, HepPar1 is frequently used as marker for HCC (4)(5)(6)(7)(8) and CK 7 and CK19 for ICC (9)(10)(11).…”

“…Currently, ultrasound sonography (US), computed tomography (CT), magnetic resonance imaging (MRI), and histopathological examination for tumor biopsy are used for diagnosis. However, distinguishing the three different primary liver tumors is often a challenging task in diagnosis, for which immunohistochemical analysis for specific antigens is a helpful tool: ·-fetoprotein (AFP) and hepatocyto paraffin 1 (HepPar1) for HCC (4)(5)(6)(7)(8) and cytokeratin (CK) 7 and CK19 for ICC (9-11).…”
