The SLITRK4-CNPY3 axis promotes liver metastasis of gastric cancer by enhancing the endocytosis and recycling of TrkB in tumour cells

Abstract: Gastric cancer (GC) is a malignant tumour with high mortality, and liver metastasis is one of the main causes of poor prognosis. SLIT-and NTRK-like family member 4 (SLITRK4) plays an important role in the nervous system, such as synapse formation. Our study aimed to explore the functional role of SLITRK4 in GC and liver metastasis. MethodsThe mRNA level of SLITRK4 was evaluated using publicly available transcriptome GEO datasets and Renji cohort. The protein level of SLITRK4 in the tissue microarray of GC was … Show more

“…The outlook for patients with GC who have distant organ metastases is typically unfavorable[ 15 ]. The liver is the primary organ that GC spreads to, with a liver metastasis rate ranging from 36% to 40%[ 16 - 20 ].…”

“…TrkB is a significant suppression target in many cancer cells [ 92 95 ], as in many other conditions driven by excessive proliferation, such as endometriosis [ 96 ]. TrkB activation enhances the survival and proliferation of cancer stem-like cells as well as increasing metastasis, epithelial-mesenchymal transition (EMT), vascular invasion and poor prognosis [ 97 100 ]. As highlighted above (see Figure 1 ), TrkB effects are complicated by the presence of TrkB-FL and TrkB-T1 on the mitochondria and/or plasma membranes, with any differential effects of TrkB isoforms and membrane site still to be determined in tumor microenvironment cells.…”

“…The signaling pathways involving tissue factor and IL-6R can mutually influence one another, thus intensifying the activation of inflammatory cells and cytokine release. [ 34 , 35 ] FABP4, integral to both fatty acid metabolism and the inflammatory response, interacts with these markers, contributing to the broader inflammatory milieu. [ 36 , 37 ]…”
