The sweet trap in tumors: aerobic glycolysis and potential targets for therapy

Abstract: Metabolic change is one of the hallmarks of tumor, which has recently attracted a great of attention. One of main metabolic characteristics of tumor cells is the high level of glycolysis even in the presence of oxygen, known as aerobic glycolysis or the Warburg effect. The energy production is much less in glycolysis pathway than that in tricarboxylic acid cycle. The molecular mechanism of a high glycolytic flux in tumor cells remains unclear. A large amount of intermediates derived from glycolytic pathway cou… Show more

“…Also, lactate production remained low when cells were exposed to 0.1 μmol/L IM for 4 weeks (Figure B). Furthermore, we observed reduced expression of HIF‐1α protein, which contributes to increased glycolysis and proliferation of cancer cells, and reduced expression of mRNA encoding glycolytic enzymes hexokinase 2 and lactate dehydrogenase A in K562‐IM3w cells (Figure C,D). Inhibiting glycolysis using 2‐deoxy‐ d ‐glucose increases the efficacy of several anti‐cancer agents; however, inhibiting glycolysis in K562 cells using 2‐deoxy‐ d ‐glucose did not increase their sensitivity to IM (Figure E), despite marked inhibition of glycolysis (Figure ).…”

“…Glycolysis is a low energy yield process but is more simple and fast. Therefore, glycolysis is exacerbated in tumor tissue and increases with tumor progression [32]. …”

“…There are many proteins participating in regulating aerobic glycolysis including glucose transporters 1 (GLUT1), lactate dehydrogenase A (LDHA), and PDH kinase (PDK) . GLUT1 facilitates the transport of glucose into cancer cells . LDHA is responsible for catalyzing the final glycolytic step that converts pyruvate into lactate .…”
