The Synergistic Effect of Cationic Moieties and GRGDSF‐Peptides in Hydrogels on Neural Stem Cell Behavior

Abstract: This article reports the behavior of embryonic neural stem cells on a hydrogel that combines cationic, non-specific cell adhesion motifs with glycine-arginine-glycine-aspartic acid-serine-phenylalanine (GRGDSF)-peptides as specific cell adhesion moieties. Therefore, three hydrogels are prepared by free radical polymerization that contains either a GRGDSF-peptide residue (P1), amino ethylmethacrylate as a cationic residue (P2), or a combination of both motifs (P3). For each gel, cross linker concentrations of 8… Show more

“…Cells were stained immunocytochemically as previously described (Czopka, von Holst, ffrench‐Constant, & Faissner, ; Kandasamy, Roll, Langenstroth, Brustle, & Faissner, ; Sallouh et al, ). After removal of the culture medium from primary OLs, cells were washed once with KRH/A (Krebs‐Ringer‐HEPES buffer pH 7.3 + 0.1% [w/v] BSA) and primary antibodies for extracellular or membrane bound epitopes (O4) were diluted in KRH/A and incubated with the cells for 30 min at RT.…”

“…While cationic charges, substrate modulus and peptides on their own are promising tools to influence the behavior of NSCs, they are very often not combined in a single hydrogel (Jaiswal et al, 2013;Macková et al, 2016). Therefore, Sallouh et al (2017) investigated the effect of both the cationic charge and the peptide sequence GRGDSF in a cross-linked hydrogel system based on poly(acrylamide). In total the effect of three different hydrogels, namely a neutral gel bearing the peptide GRGDSF, a gel with cationic charges based on 2-aminoethylmethacrylate and a gel bearing both cationic charges and the peptide GRGDSF, on the behavior of NSCs were analyzed (see Figure 2).…”
