The Th17 Pathway in Vascular Inflammation: Culprit or Consort?

Abstract: The involvement of IL-17A in autoimmune and inflammatory diseases has prompted the development of therapeutic strategies to block the Th17 pathway. Promising results came from their use in psoriasis and in ankylosing spondylitis. IL-17A acts on various cell types and has both local and systemic effects. Considering the premature mortality observed during chronic inflammatory diseases, IL-17A action on vascular cells was studied. Both in vitro and in vivo results suggest that this cytokine favors inflammation, … Show more

“…87,93,94 Exacerbated Th17 activation may induce and contribute to the onset or progression of autoimmune disorders such as T1D 95 and vascular inflammation. 96 The balance between Treg/ Th17 cells is critical and is responsible for generation of pro-inflammatory cytokines such as IL-17 production, which are known to facilitate the development of diabetic retinopathy. [97][98][99][100] Thus, collectively our study supports that retinopathy in human subjects with T1D is associated with a dysregulated systemic RAS and profound gut permeability defects which triggered activation of components of both the adaptive (B-cells, Treg/Th17, DCs) and innate (granulopoiesis) immune response and changes in gut derived ILC1s.…”

“…Despite a pivotal role, the Th2 response alone cannot fully explain other EGPA manifestations, such as granulomas and vascular inflammation. An increased frequency in IL-17-producing CD4 þ T cells (Th17) has been described in EGPA compared to healthy or asthmatic controls and during active disease compared to inactive EGPA [14]. Moreover, a defective activity of regulatory T cells (Treg), which exert a suppressive effect on Th17 cells, has been described during active EGPA.…”

“…This is a critical element of immune regulation since excessive IL‐6 production increases Th17 cells important in mediation of allograft rejection. We now know that Th17 cells contribute to allograft injury, suppress Treg cell development and stimulate profibrotic cytokines resulting in progressive declines in renal function and graft loss 18 …”
