The Therapeutic Prospects of Targeting IL-1R1 for the Modulation of Neuroinflammation in Central Nervous System Disorders

Abstract: The interleukin-1 receptor type 1 (IL-1R1) holds pivotal roles in the immune system, as it is positioned at the “epicenter” of the inflammatory signaling networks. Increased levels of the cytokine IL-1 are a recognized feature of the immune response in the central nervous system (CNS) during injury and disease, i.e., neuroinflammation. Despite IL-1/IL-1R1 signaling within the CNS having been the subject of several studies, the roles of IL-1R1 in the CNS cellular milieu still cause controversy. Without much dou… Show more

“…The ORAI2 gene is a decisive factor in classifying AD samples as 0 and the gene 7981324 as 1 on frontal AD and non-AD samples, respectively. Figure 7 shows the SHAP interpretation on hippocampal AD and non-AD samples, identifying 8,015,796 and IL1RL2 (Luís et al 2022 ) as the top contributors to the predictions. The ORAI2 gene is found to have higher significance in a temporal dataset of both classes and stands on top of the other genes, as depicted in Fig.…”

“…S100A9 activates NF-κB signaling and has been shown to aggravate neuroinflammation in a model of subarachnoid hemorrhage 108 and Alzheimer’s disease 109 and has been implicated as an integral component of inflammation along the gut-brain axis in the DSS model 15 . Cyclin-dependent kinase inhibitor p21, encoded by Cdkn1a , appears to be an inflammatory response element and may be a marker of DNA damage 110,111 , and an upregulation of the IL-1 receptor type 1, encoded by Il1r1 , has been associated with pathogenesis in several neurodegenerative diseases 112 . In general, the colitis-induced neuroimmune response is complex, and further work will be needed to unravel this complexity especially amidst LC injury.…”
