The transcriptional repressor JHDM3A demethylates trimethyl histone H3 lysine 9 and lysine 36

Abstract: Post-translational modification of chromatin has profound effects on many biological processes including transcriptional regulation, heterochromatin organization, and X-chromosome inactivation. Recent studies indicate that methylation on specific histone lysine (K) residues participates in many of these processes. Lysine methylation occurs in three distinct states, having either one (me1), two (me2) or three (me3) methyl groups attached to the amine group of the lysine side chain. These differences in modifica… Show more

“…We also provide important evidence that Ring1B/Rnf2 forms distinct complexes with the histone demethylases LSD1/Aof2 and Fbxl10/ Jhdm1B, which demethylate H3 Lys-4 and H3 Lys-36, respectively (63,64). Whereas methylation of these histone H3 lysine residues has been associated with transcriptional activation (65), there is recent evidence implicating histone demethylases in repression (66). Taken together, Ring1B/Rnf2 complexes have the capacity to ubiquitinate H2A Lys-119, to methylate H3 Lys-9 (Ehmt1-Ehmt2 and maybe Setdb1), and to demethylate H3 Lys-4 (Lsd1/Aof2) and/or H3 Lys-36 (Fbxl10/Jhdm1B) most likely in combinations of (distinct and/or overlapping) activities and presumably binding to different gene target subsets.…”

“…JMJD2A has histone demethylase activity for H3 K9 and K36 (Klose et al, 2006;Whetstine et al, 2006). Its double tudor domain binds methylated H3 K4 and H4 K20 (Huang et al, 2006).…”

“…Although JMJD1 family proteins are specific for H3K9 mono-and dimethylation, 15 the members of JMJD2 family demethylate tri-and dimethylated H3K9 and H3K36 (Table 1). 2,14,32,33 Nevertheless, little is known about the implication of JMJD1 in tumorigenesis. Frequent deletion of 5q31 locating JMJD1B gene or reduced expression of JMJD1C in various malignancies suggests possible roles of JMJD1 proteins in tumor suppression.…”
