The Urokinase Receptor Homolog Haldisin Is a Novel Differentiation Marker of Stratum Granulosum in Squamous Epithelia

Abstract: Several members of the Ly-6/uPAR (LU)-protein domain family are differentially expressed in human squamous epithelia. In some cases, they even play important roles in maintaining skin homeostasis, as exemplified by the secreted single domain member, SLURP-1, the deficiency of which is associated with the development of palmoplantar hyperkeratosis in the congenital skin disorder Mal de Meleda. In the present study, we have characterized a new member of the LU-protein domain family, which we find to be predomina… Show more

“…Consistent with previous reports, restricted expression in stratum granulosum was found for filaggrin (FLG) and FLG2 (data not shown); Ly6/PLAUR domain-containing protein 5 (LYPD5) (Gardsvoll et al 2013); keratinocyte proline-rich protein (KPRP; Fig. 3O) (Kong et al 2003); premature ovarian failure 1B protein (POF1B; Fig.…”

“…19,23,24 In this system, the GPIHBP1 proteins contained an amino-terminal uPAR tag (detectable with antibody R24) and sequences from the carboxyl terminus of mouse GPIHBP1 (detectable with antibody 11A12). Under nonreducing conditions, 48.8 ± 0.02% ( n = 20 experiments) of the wild-type GPIHBP1 secreted by Drosophila S2 cells was monomeric, as judged by western blots with antibody 11A12 (Figure 1B).…”

“…In the present study, we have examined the expression patterns of C4.4A and Haldisin, two newly discovered squamous differentiation markers (Gårdsvoll et al 2013;Kriegbaum et al 2011), in a collection of skin diseases. Whereas we found no dramatic alterations in their expression signatures in the various blistering lesions covering most of the strata in the squamous epithelium, or in the lesions with dysregulated differentiation, we did observe attenuation in C4.4A expression in the lesion core of invasive squamous cell carcinomas and keratoacanthomas and a recovery of high expression in the deeply invasive fronts.…”

“…Recombinant human C4.4A and Haldisin as well as rabbit anti-C4.4A and anti-Haldisin polyclonal antibodies (pAb) were generated and purified as previously described (Gårdsvoll et al 2013;Hansen et al 2004). Rabbit anticytokeratin (CK) 5 pAb (ab24647), mouse anti-CK14 monoclonal antibody (mAb) (ab7800) and rabbit anti-loricrin pAb (ab24722) were purchased from Abcam (Cambridge, UK).…”
