The use of fluorescein-conjugated Bandeiraea simplicifolia B4-isolectin as a histochemical reagent for the detection of α-d-galactopyranosyl groups

Abstract: The Bandeiraea simplicifolia B,-isolectin, which combines specifically with a-o-galactopyranosyl groups, has been conjugated with fluorescein isothiocyanate and demonstrated to be a reliable histochemical probe for the detection of these groups in normal tissues of mouse, rabbit, rat and man. Specificity of binding of the fluorescein-conjugated B. simplicifolia B,-isolectin to native cryostat tissue sections was demonstrated in two ways:1. The hapten inhibitor methyl a-Bgalactopyranoside prevented the binding … Show more

“…Fluoresceinated-B4 isolectin was shown to bind in a specific manner to basement membrane structures in a variety of murine tissues [8]. We now believe that laminin is the co~astituent of basement membrane to which the B4 isolectin binds.…”

“…Of the a-galactosidase activity initially present in the digestion mixture 70% remained after 94 h incubation. Relative to the ~-galactosidase activity, other glycosidase activities were <-0.03% as determined with p-nitrophenylglycoside substrates [8]. Control mixtures lacked either the ot-galactosidase or the laminin.…”

“…The B-subunit shows a high degree of specificity toward α-D-Galp but not α-D-GalNAcp groups. BSL-B4, homo-tetramer of B-subunits, exhibited binding to the glomerular and peritubular basement membrane in frozen sections of mouse and rabbit kidneys, but not in those of humans [28]. Biotinylated BSL-I used in the present study is a mixture of five isolectins, binds mainly with α-D-Galp groups [19].…”
