The use of high-dose estrogens for the treatment of breast cancer

Abstract: Estrogens are known to stimulate the growth of breast cancer but they are also an effective treatment for this disease (this has been termed the 'estrogen paradox'). The fact that estrogens can be an effective treatment for breast cancer is something that has almost been forgotten, whereas the fear for estrogens remains. This paper reviews the use of estrogens for the treatment of breast cancer and identifies possible applications. The data summarised in this review demonstrate that high-dose estrogens are eff… Show more

“…Oral contraceptives (OCs) were developed in the early 1960s comprising predominantly a steroidal estrogenic compound ethinylestradiol (EE), and later, a synthetic 17-beta estradiol (SE2) also was used as an OC component [66]. OCs comprising low doses of synthetic estrogens, apparently worked well in women, however, in certain cases they evoked unforeseeable complications, such as venous thromboembolism, stroke and myocardial infarct suggesting severe clotting disorders [67,68].…”

“…In fact, its role is pivotal in regulating reproduction in humans; for this reason, several oral contraceptives contain the synthetic estrogen ethinylestradiol (Kuhl, 2005;Evans and Sutton, 2015). Moreover, estradiol is also used in therapies to treat prostate and breast cancer (Ali Shah, 2015;Coelingh Bennink et al, 2017). In 2016, more than 13 million prescriptions of estradiol were made allowing for the 59th most prescribed medication in the United States.…”
