The voltammetric determination of peroxynitrite at a mercury film electrode

Abstract: This report describes the direct voltammetric detection of peroxynitrite (PN, ONOO-), an important analyte in many physiol. biochem. pathways, at a mercury film electrode (MFE) at alk. pH. The voltammetric response of PN is obsd. as a cathodic inverted peak when the potential is swept in the oxidative direction at ~-0.1 V vs. a Ag/AgCl/1 M KCl ref. electrode. An mechanism is proposed to explain this behavior involving the initial formation of a thin film of Hg(I) oxide on the electrode which is then oxidized b… Show more

“…Peroxynitrite is a strong oxidant [E′ o 1.4 v for ONOO − /NO − 3 (Zakharova et al, 2007)]. However, the many of its biological effects can be due to the reactions of its decomposition product, NO 2 (Pacher et al, 2007) [E′ o 0.8 v for NO 2 /NO − 3 (Standard electrode potential data)].…”

“…Hb is used as the mimetic enzyme for substituting horseradish peroxidase for two reasons: (1) for the purpose of the mildness of reaction conditions and the minimization of cost and (2) Hb is a kind of nature protein in human body and it is much likely to coexist with peroxynitrite in cell or organism (Liang et al 2005). Other peroxynitrite determination methods are direct voltammetric detection at a mercury film electrode at alkaline pH (Zakharova et al 2007) and luminol-enhanced chemiluminescence methods (Magalhaes et al 2008).…”

“…62 While sufficiently stable for 30 to 60 minutes in alkaline solutions (pH z 10), 17 peroxynitrite decomposes in less than 1 s at neutral pH. 65 Since using alkaline buffer PON solutions is customary in electrochemical sensing methods, 66,67 the following experiments were conducted in CAPS buffer (pH 10).
