The β Subunit of the Sec61p Endoplasmic Reticulum Translocon Interacts with the Exocyst Complex in Saccharomyces cerevisiae

Abstract: The exocyst is a conserved protein complex proposed to mediate vesicle tethering at the plasma membrane. Previously, we identified SEB1/SBH1, encoding the ␤ subunit of the Sec61p ER translocation complex, as a multicopy suppressor of the sec15-1 mutant, defective for one subunit of the exocyst complex. Here we show the functional and physical interaction between components of endoplasmic reticulum translocon and the exocytosis machinery. We show that overexpression of SEB1 suppresses the growth defect in all e… Show more

“…The recent discovery that the vesicle-tethering exocyst has a conserved role in communicating with the ER-translocon complex (Lipschutz et al, 2003;Toikkanen et al, 2003) lends credence to such an idea. Furthermore, upon an ExPASy proteomic server (http://br.expasy.org/) search for motifs on IQGAP1, we identified an ER membrane-retention signal (KFYG) on the extreme Cterminus.…”

“…Anti-Sec9p-CT antibodies have been described previously (Brennwald et al, 1994). AntiSec4p (R232), Sec15p (R233), Sso1/2p (K8), Sso1p (K6916), Sso2p (K6906), Mso1p (R285), tubulin, and Ady3p antibodies have been described previously (Aalto et al, 1997;Moreno-Borchart et al, 2001;Jantti et al, 2002;Toikkanen et al, 2003). Anti-Sec8p antibody was obtained from Wei Guo (University of Pennsylvania, Philadelphia, PA).…”

“…23) The resulting Sec61β/exocyst complexes upregulate post-transcriptional synthesis and delivery of secretory and basolateral membrane proteins. [24][25][26] Given that claudins are basolateral membrane proteins, they may also be subjected to Sec61β-mediated upregulation.…”
