Theabrownin Isolated from Pu-Erh Tea Enhances the Innate Immune and Anti-Inflammatory Effects of RAW264.7 Macrophages via the TLR2/4-Mediated Signaling Pathway

Abstract: Theabrownin (TB) is a tea pigment extracted from Pu-erh Tea. The effects of TB on innate immunity and inflammation are not well understood. Herein, the effects of TB on innate immunity are investigated using RAW264.7 macrophages. We found that TB promoted the proliferation of RAW264.7 macrophages, altered their morphology, enhanced their pinocytic and phagocytic ability, and significantly increased their secretion of nitric oxide (NO) and cytokines, all of which enhanced the immune response. Additionally, TB i… Show more

“…32,33 It possesses antioxidant properties that combat harmful free radicals, reducing oxidative stress and inflammation, which may delay neurodegenerative processes. 34–36 These findings encouraged us to further explore the active constituents of Pu-erh tea that inhibit COMT activity and delve deeper into its mechanism of COMT inhibition.…”

“…TB is a microbial fermented polyphenol found in dark tea like Pu-erh tea and Fu Brick tea [ 73 , 74 ]. The health benefits of TB include preventing tumors, improving immunity, anti-inflammation, alleviating obesity and metabolic syndromes, and ability to improve gut microbiota [ 73 , 74 , 75 , 76 , 77 , 78 ]. TB demonstrated anti-tumorigenesis effects on CRC in the AOM/DSS model [ 33 ].…”

“…In the analysis of the therapeutic effect of six kinds of tea on the liver injury induced by carbon tetrachloromethane (CCl4) in mice, dark tea can inhibit the NF-κB pathway to be anti-inflammatory and reduce liver injury, while green tea, yellow tea, oolong tea, and white tea have no such effect [ 43 ]. TB extracted from Pu’er tea regulates the immunity of RAW264.7 macrophages and inhibits inflammation by inhibiting the NF-κB/MAPK/PI3K-AKT signaling pathway [ 44 ]. Dark tea has also been shown to alleviate colitis induced by dextran sodium sulfate and reduce the expression level of inflammatory factors, mainly by regulating the NF-κB and HIF-1α signaling pathways, regulating gut bacteria, and enhancing the synthesis of short-chain fatty acids such as butyrate [ 45 ].…”
